Allison Janney nació en Boston pero creció en Dayton (Ohio), hija de una exactriz y ama de casa llamada Macy y de un promotor inmobiliario y músico de jazz llamado Jervis Spencer Janney. Tiene dos hermanos mayores llamados Hay y Hal. Estudió en la escuela Miami Valley School de Dayton, de la que fue nombrada alumna distinguida en 2004.
