He was born 9 December 1761 in Spring Gardens, London, the second surviving son of Thomas Penn and his wife, Lady Juliana Fermor, fourth daughter of Thomas, first Earl of Pomfret.

Granville Penn (1761-1844) was an amateur textual critic, who intended to revise the English translation of the New Testament, and brought in evidence from Codex Vaticanus (B03) in order to support his argument.
John Penn had Pennsylvania castle built on the island, and after his death the castle and its contents were inherited by Granville. Granville was also the principle heir of Philadelphia Hannah Freame [1740/1741-1826], granddaughter of William Penn [1644-1718], the founder of Pennsylvania.
