È celebre soprattutto per aver interpretato il personaggio di Tarzan, in cinque pellicole distribuite dalla RKO Radio Pictures tra il 1949 e il 1953, e il personaggio di Old Shatterhand, alter ego dello scrittore tedesco Karl May, in una serie di film realizzati in Germania tra il 1962 e il 1968.
In Tarzan's Magic Fountain (1949), Barker became the tenth official Tarzan of the movies. His blond, handsome, and intelligent appearance, as well as his athletic 6'4" frame, helped make him popular in the role Johnny Weissmuller had made his own for 16 years.
