Hamlin was the son of Cyrus Hamlin, a physician, sheriff, and farmer, and Anna Livermore. After practicing law, he entered politics as an antislavery Jacksonian.

Hamlin originally entered the U.S. Senate as a Democrat but he was one of the few Democrats to vote against the Kansas-Nebraska Act because he did not want slavery to spread into the territories. He renounced his affiliation with the Democratic Party in 1856 and served in the Senate until 1857 when he resigned to run for governor.
Hannibal Hamlin owed his classical name to the influence of his grandfather, who loved the great military figures of ancient history. Tall, with piercing black eyes and olive-colored skin, the courteous and affable Hamlin proved to be a natural politician.
