Hamlin originally entered the U.S. Senate as a Democrat but he was one of the few Democrats to vote against the Kansas-Nebraska Act because he did not want slavery to spread into the territories. He renounced his affiliation with the Democratic Party in 1856 and served in the Senate until 1857 when he resigned to run for governor.
