Domhnall Gleeson naît à Dublin en Irlande de l' acteur Brendan Gleeson 3 et de Mary Gleeson (née Weldon). Il a trois frères cadets : Fergus, Brian, qui est aussi acteur et Rúairí. Il a reçu le diplôme de bachelor of arts degree dans les arts des médias au Dublin Institute of Technology 3 .
