James Hamilton è nato il 4 luglio 1934 ed è il figlio di James Hamilton, IV duca di Abercorn, e Lady Mary Crichton, figlia del tenente colonnello Henry William Crichton, visconte Crichton ( 1872 - 1914) e nipote di John Crichton, IV conte Erne . Ha studiato all' Eton College e al Royal Agricultural College di Cirencester.

He became the 5th Duke of Abercorn in the Peerage of Ireland on the death of his father, the 4th Duke, in 1979.
